Chronic intraperitoneal administration of AOD-9604 to obese mice for 14 days reduced body weight and body fat, an effect the authors associated with increased expression of beta-3 adrenergic receptor RNA, a pathway involved in fat breakdown; the effect was absent in beta-3 adrenergic receptor knock-out mice, though an acute dose of AOD-9604 still increased energy expenditure and fat oxidation in those knock-out mice. A separate study using mini-osmotic pumps to deliver AOD-9604 to obese and lean mice over 14 days found increased fat oxidation and reduced weight gain without the hyperglycemia or reduced insulin secretion seen with full-length growth hormone, and in vitro assays showed AOD-9604 did not bind the growth hormone receptor or stimulate cell proliferation the way full-length growth hormone did.
