Ipamorelin is a synthetic pentapeptide described in the pharmacology literature as a selective growth hormone secretagogue: in animal and in vitro work it released growth hormone with potency similar to a reference peptide (GHRP-6) while not significantly raising ACTH or cortisol, unlike some other secretagogues tested in the same study [1]. A human pharmacokinetic and pharmacodynamic dosing study in healthy male volunteers examined dose-proportional growth hormone release after intravenous infusion [2]. Ipamorelin has no FDA-approved indication and is sold only as a research chemical.

Selectivity for GH over ACTH and cortisol in animal and in vitro models

In rat pituitary cells, anaesthetized rats and conscious swine, ipamorelin released growth hormone with potency and efficacy comparable to GHRP-6; in swine, none of the growth hormone secretagogues tested (including ipamorelin) altered FSH, LH, prolactin or TSH levels, and ipamorelin, unlike GHRP-6 and GHRP-2, did not raise ACTH or cortisol above levels seen with GHRH stimulation alone, even at doses more than 200-fold above its effective dose for GH release. Human dose-response study

A pharmacokinetic-pharmacodynamic study in healthy male volunteers used an ascending intravenous infusion design across several dose levels and reported dose-proportional pharmacokinetics with a short terminal half-life of about two hours, alongside measured growth hormone responses. [2]

What does dosing evidence actually support?

The human data available comes from a controlled infusion pharmacokinetic study using intravenous administration in a small cohort of healthy male volunteers, designed to characterize dose-response and clearance rather than to establish a therapeutic or wellness dosing protocol [2]. That study did not use subcutaneous injection at the doses or frequency commonly discussed in non-clinical settings, so it describes measured pharmacology, not a validated dosing regimen for any other route or population.
