What is MOTS-c?

MOTS-c is a naturally occurring 16-amino-acid mitochondrial-derived peptide, distinct from a synthesized research reagent sold under the same name. Studies use it in mouse and cell models or measure endogenous serum levels in patients; a manufactured vial is a research chemical, not the same regulatory entity as the endogenous signaling peptide characterized in these papers.

What the studies reported

Circulating levels reduced in a metabolic disease population

A study found reduced serum and skeletal-muscle MOTS-c in women with polycystic ovary syndrome compared to controls, and reported that this reduction correlated with markers of mitochondrial dysfunction. [1]

Animal model shows inflammasome suppression

In a diabetic rat model, MOTS-c treatment suppressed systemic and cardiac NLRP3 inflammasome activation, a rodent finding proposed as a mechanism behind MOTS-c's studied cardioprotective effects in that model. [2]

Exercise raised circulating MOTS-c in one human study

A study measuring circulating mitochondrial-derived microproteins in people with cerebral palsy found that MOTS-c and related microproteins increased in blood after a single acute bout of endurance exercise, a single human study rather than a broader literature consensus. [3]

What does dosing evidence actually support?

No completed human clinical trial administering exogenous MOTS-c was identified in the sources reviewed. Available human data are observational, correlating endogenous serum MOTS-c concentrations with conditions such as polycystic ovary syndrome, myocardial infarction, and exercise status. Dosing information that exists comes exclusively from rodent studies (diabetic and cachexia models) using injected peptide at researcher-selected doses for specific experimental endpoints, and these figures do not constitute a validated human protocol of any kind.

A study dose is not a personal dose. Population, route, formulation and supervision are part of the result. Online community schedules are not treated here as established clinical protocols.

What not to overlook

  • No human interventional dosing trial was found; do not extrapolate rodent research doses to any human use.
  • Observational human studies measured endogenous peptide levels, not effects of an administered product.
  • A manufactured research vial is a synthesized peptide reagent, not verified equivalent to the endogenous signaling peptide studied in these papers.
  • Cardiac and metabolic disease associations reported so far are correlational, not evidence of therapeutic benefit from supplementation.

Names are not interchangeable

Products are typically sold as lyophilized peptide powder in single-use research vials for laboratory use, distinct from any pharmaceutical formulation, since no approved MOTS-c drug product exists. No blend ratio applies to single-component MOTS-c vials; identity, purity, and intended research-only use should be verified against the specific vendor's certificate of analysis rather than assumed.

Follow validated product instructions
