What is NAD+?

NAD+ is a small-molecule coenzyme, chemically unrelated to peptides, present naturally in all cells. Human studies vary widely by what was actually administered (NAD+ itself, nicotinamide riboside, or nicotinamide mononucleotide), by what tissue compartment was measured (whole blood versus plasma), and by route (oral, sublingual, intravenous), so results are not interchangeable between formulations.

What does dosing evidence actually support?

Human trials reviewed used varied routes (oral, sublingual, intravenous) and forms (NAD+, nicotinamide riboside, nicotinamide mononucleotide) in specific study populations such as older adults with cognitive impairment or Friedreich's ataxia patients, generally over weeks to months, with study-selected doses that are not standardized across the field. These figures describe what was tested in each named study, not a general recommendation, and results should not be assumed to transfer between NAD+ and its precursor molecules or between administration routes.

A study dose is not a personal dose. Population, route, formulation and supervision are part of the result. Online community schedules are not treated here as established clinical protocols.

What not to overlook

  • NAD+, nicotinamide riboside, and nicotinamide mononucleotide are related but distinct molecules studied separately; findings for one should not be assumed to apply to another.
  • Intravenous and oral/sublingual routes produced different tolerability and blood-level findings in the studies reviewed and are not interchangeable.
  • Most rigorous randomized trials located targeted specific disease populations, not general healthy-adult supplementation use.
  • Long-term safety data in healthy adults remain limited in the sources reviewed.

Names are not interchangeable

Commercial research NAD+ is typically sold as a lyophilized powder for reconstitution, separate from the oral precursor supplement products (nicotinamide riboside, nicotinamide mononucleotide) discussed in most clinical literature. Studies cited here tested specific pharmaceutical or supplement-grade preparations; a research vial's formulation, purity, and route are not established as equivalent to any studied product without independent verification.

Follow validated product instructions

A single water volume, refrigerator temperature or expiry window cannot be assumed for every compound and formulation. Use the exact manufacturer documentation and a validated laboratory procedure. If stability, solvent compatibility or microbiological quality is not documented, those properties remain unverified.
